چکیده

Glycopeptide-resistant enterococci (GRE) can carry more than one VanA element (3, 5), but we are unaware of previous evidence for separate transfer of such elements. Enterococcus faecalis JS3B was examined as one of 46 GRE isolated from fecal screens of a hematology patient at Addenbrooke’s Hospital, Cambridge, United Kingdom (7). Overlapping PCR (1) showed that this strain contained a group H element (11). VanA resistance appeared to be associated with a 35-MDa plasmid, but Palepou et al. (6) suggested that group H elements reside on the chromosome. Strain JS3B was therefore investigated for carriage of multiple VanA elements. Transfer of vancomycin resistance was performed by crossstreak conjugation (10). The recipient strains were E. faecalis JH2-2 and E. faecium GE-1, which are resistant to fusidic acid and rifampin and lack pheromone response genes prgA and prgB (see below). After incubation at 37°C for 72 h, the transconjugants were selected on brain heart infusion agar

vanA in Enterococcus faecium, Enterococcus faecalis, and Enterococcus casseliflavus detected in French cattle.

The goal of this study was to assess the presence of enterococci species presenting van-mediated glycopeptide resistance in French cattle. Fecal samples were collected from healthy and sick animals, and enterococci were screened for vancomycin resistance. Vancomycin resistance was principally encountered in Enterococcus gallinarum and Enterococcus casseliflavus strains. However, glycopeptide re...

Molecular characterization of enterococci harboring genotype and phenotype incongruence related to glycopeptide resistance isolated in Brazilian hospitals.

Three Enterococcus faecalis and one Enterococcus faecium strains were characterized by plasmid profile, pulsed-field gel electrophoresis (PFGE) and determination of antimicrobial minimal inhibitory concentrations. VanA elements were characterized by Long PCR, overlapping PCR and DNA sequencing. Enterococcal strains showed resistance to vancomycin and harbored the vanA gene, and three these were...

Restoration of vancomycin susceptibility in Enterococcus faecalis by antiresistance determinant gene transfer.

We assessed the ability of gene transfer to reverse vancomycin resistance in class A (VanA) glycopeptide-resistant Enterococcus faecalis. Recombinant shuttle vectors containing a vanH promoter-vanA antisense gene cassette fully restored vancomycin susceptibility through a combined transcriptional activator binding domain decoy and inducible vanA antisense RNA effect.

Heterologous expression of the enterococcal vanA operon in methicillin-resistant Staphylococcus aureus.

Two methicillin- and vancomycin-resistant Staphylococcus aureus strains, MI-VRSA and PA-VRSA, and Enterococcus faecalis DMC83006B, considered to be the potential donor of glycopeptide resistance to MI-VRSA, were studied. MI-VRSA is highly resistant to both glycopeptides, whereas PA-VRSA displays low-level resistance to vancomycin and reduced susceptibility to teicoplanin. We have analyzed the e...

Presence of glycopeptide-encoding plasmids in enterococcal isolates from food and humans in Denmark.

Enterococci and especially glycopeptides-resistant enterococci (GRE) are a growing concern due to their ability to cause infections in hospitals. Transmission of antimicrobial resistance between reservoirs such as animals, meat, and humans are in most cases linked to transmission of mobile genetic elements (MGE) such as plasmids and transposons. Presence of MGE was tested in all GRE isolated fr...
